Engineering the Salmonella SPI-1 System for Heterologous Protein Production

Abstract
We are engineering the multi-gene prokaryotic type III secretion system for biotechnological applications. The Salmonella system pumps proteins from the cytoplasm, through both membranes, to the extracellular environment. Although the system is naturally utilized for the secretion of virulence proteins, we have shown previously that it can be exploited to deliver heterologous proteins such as spider silk monomers to the growth media, and are now developing methods to optimize this production. Specifically, we have examined and manipulated the natural regulatation of the system to increase the levels of secreted protein.
